There seems to be some demand for a GLYX-13 discussion (now known as Rapastinel) so I would like to start one here.

NEW INFORMATION HAS COME OUT THAT BECAUSE KETAMINE IS SO DANGEROUS AND CONTROLLED IT WILL NEVER BE AVAILABLE. THE BEAUTY IS THAT IT OPENED UP THIS WHOLE NEW ARE OF BRAIN RESEARCH CALLED THE GLUTAMATE SYSTEM AND ITS DISFUNCTION BEING A MAJOR PLAYER IN DEPRESSION.

There is actually a doctor administering ketamine intravenously for depression in my city, I've been told. His name is Dr. David Faifel, and he works at UCSD Medical Center.

All-sorry I just can just type faster. Time will tell. Ketamine is Not sanctioned by the National Institute of Mental Health. Nor is anyone trying to get approval from the FDA for an Intravenous Anesthetic for Depression. Nor are there anymore trials. Dangerous side effects,
Daisy, are you in LA? me too. Who is your regular Psychiatrist?
Where are you doing the GLYX-13 trial?
I got rejected for Breast Cancer. I cannot get into any trials.....
I called USC way back. They only use it for pain for a specific type of disease for pain treatment. That is what they told me on the phone. The MD that was doing it at UCLA, where I go, stopped.....

As I stated before, I'm in a trial of AZD6765. I don't live in LA, but you can check to see which trials are being conducted there by going to clinicaltrials.gov

Daisy- what are the specifics of the AZD6765 trial? You mentioned that there was a high-dose and a low-dose group. Is there a placebo group? Typically these trials will always have a placebo group.

Yes, there is a placebo. I know that I'm not in that group though, because of the way I'm feeling.

I can post which dose I'm on if/when Astra Zeneca unblinds the trial, if anyone wants me to.

I went to the trial place for GLYX-13 and they were about to interview me to be in the trial. But then, the bombshell...2 week washout period for the Wellbutrin, I can't take the chance of becoming so depressed I will need to be hospitalized. So I decided not to participate. ): The woman there said she had seen some incredible results for people, I know this, I have experienced 'the miracle'. I just can't go back to *the hole*, barely hanging on now. But very interested to hear of others experiences. Best of luck to everyone!

Paula,

AZD6765 is being studied only as an adjunct medication, so you actually have to be on an A/D to qualify for a clinical trial.

A doctor prescribed Namenda for my depression last year for the reasons you stated, but I was afraid to take it for fear of the side effects I'd read about it.

Regarding your migraines, have you tried the SNRI Remeron (mirtazapine)? I used to get migraines too, and they pretty much disappeared after I started taking Remeron. It's the only a/d I can tolerate, and I still take it. I realize that everyone's different, though.

Yes, I tried remeron. It didn't help the migraines but made it impossible for me to stop eating :) I haven't met a SSRI or SNRI yet that I can tolerate. Thanks for the thought tho!

Daisy, after only 3 days on Namenda, it appears to be a migraine TRIGGER for me all. Either that or it's the weather. I'm going to stop taking it and maybe give it another try when the weather clears up, if I can stand the thought of it.

I was curious what side effects you had when you tried it? I had 3 days of unrelenting migraines almost from the minute I took the first pill. Very disappointing to say the least, because I was hoping it would help my anxiety. If anyone else has experience with this, I would appreciate hearing (tho maybe I should switch to the Namenda page - if there is one - arrrrggghhhh)

Hey Catpower,

I filled the Rx for Namenda but didn't take it. I forget what I read about the side effects that decided me against it. I wouldn't be surprised if that's what's causing your migraines.

I'm still very pleasantly surprised at how few side effects I've experienced from the AZD6765 infusions compared to all the pills I've tried. I'm having my next-to-last infusion on Wednesday.

Sorry about the weather you're experiencing. I'm originally from NYC, so I know from humidity. Live in San Diego now, which is much more bearable...I've met people who do suffer headaches when the barometer moves in one direction or the other.

Hi Daisy

If it's not too much to ask, could you tell us a bit more about how the treatment works for you in terms of attention, memory, cognition, energy and emotional depth? I too have quit SSRIs because of terrible side effects and I am awaiting this new drug class. SSRIs made me have dampened feelings and killed my libido. Have you noticed anything similar with AZD6765??

Hi grouper,

What I've noticed is fatigue, especially the day after a treatment (like today), but it feels very different from my normal, depression-induced fatigue. The other big side effect, at least for me, has been increased appetite, which has led to weight gain of about 3 or 4 pounds. That may be because I'm also taking Remeron, which is notorious for increased appetite; I think maybe the two drugs together are having some sort of synergistic effect on my appetite.

A study doc told me yesterday that other study patients are reporting increased energy and decreased appetite, so you know, like any other drug, I think this one's side effects are going to vary from person.

The only other noticeable side effect for me was some flat affect initially, but that seemed to go away fairly quickly. I have noticed some improvement in memory.

I wish the drug were more energizing for me, but that could be dose-related--I still don't know which dose I'm on. Also, it could be that another of these drugs, like GLYX-13, will help with that (at least that's what I'm hoping).

Overall what I noticed, at least initially, was feeling more content, more level, less anxious, and finding it easier to delve into projects. I'm noticing it less now, possibly because I've tapered down from infusions three times a week to once every two weeks currently.

My opinion from having participated in this trial is that they're definitely on to something with this class of drugs, but that they're going to need fine tuning. In terms of side effects, though, I'll take them over the SSRIs any day.

Sounds like this drug is going to get to phase III after all. I 'd change a day of fatigue for low libido any time.

There's been a lot of talk about an open-label, extended study of this drug, so if you're thinking about trying it, you may have an opportunity at that point.

@ Daisy- where are they doing the study for AD???. I would be very interested in trying that. I am pretty well stabilized on my multiple-med regime, but the effects of Ketamine infusions when I was in that study last year were amazing! I felt like a new, improved me. Although did have some fatigue which was minor after the infusions. It was well worth it! Let me know. Thanks! Be well.

Paula, you tried ketamine? Did you experience any of the psycho-mimetic side effects that I've heard about? There's supposedly a doctor here in San Diego, a Dr. David Faifel, who administers ketamine infusions to treat depression. Not sure if it's covered by insurance, though...

I'm participating in the clinical trial of AZD6765 in San Diego. It's closed now, but I'm sure that if they begin a new trial, it'll be conducted in cities across the U.S. Best way to find out is to search it on clinicaltrials.gov
